Assay Procedure (Ref. No. FGAP058) - Trypsinogen

To determine the potential activity of the inactive zymogen, a solution of the preparation must be activated and the activity then measured by the metod of assay for trypsin.

The method of assay is that suggested by Schwert & Takenaka in which N-benzoyl-L-arginine ethyl ester (BAEE) is hydrolysed at the ester linkage causing an increase of absorbance measured at 253nm and 25^oC.

That amount of activated zymogen causing an increase in absorbance at 253nm of 0.003 per minute at 25^oC resulting from the hydrolysis of BAEE under the specified conditions..

Native activity is determined by measuring activity of trypsin in trypsinogen.
