Assay Procedure (Ref. No. FGAP058) - Trypsinogen

To determine the potential activity of the inactive zymogen, a solution of the preparation must be activated and the activity then measured by the metod of assay for trypsin.

The method of assay is that suggested by Schwert & Takenaka in which N-benzoyl-L-arginine ethyl ester (BAEE) is hydrolysed at the ester linkage causing an increase of absorbance measured at 253nm and 25oC.

Unit Definition

That amount of activated zymogen causing an increase in absorbance at 253nm of 0.003 per minute at 25oC resulting from the hydrolysis of BAEE under the specified conditions..

Native activity is determined by measuring activity of trypsin in trypsinogen.
